Will a PCI Express 2.0 x16 card work in a PCI Express x16 slot? Also how cani tell in my motherboard supports PCIE 2.0?

PCIe 2.0 will work in a regular slot, it just negotiates the maximum available speed

As steevo stated above, the PCIe 2.0 x16 card will work in any motherboard with a normal PCIe x16 slot.

The only motherboards (so far) supporting PCI-E 2.0 are boards with Intel's X38/48 chipset, AMD's RD770/780 and nVIDIA's nForce 7xx.
